DNA, also known as the ‘blue print’ of life, encodes the instructions for the development and function of the vast majority of organisms on our planet. Despite its importance, DNA is intrinsically unstable and is prone to breakage and chemical modifications. If left unrepaired DNA damage can giving rise to mutations, which alter the coding sequence. To counteract the deleterious effects of DNA damage, cells have evolved specialized DNA repair mechanisms. Insights into DNA repair processes and their control has firmly established the importance of DNA repair in ageing and cancer. Professional antigen-presenting cells (APC) such as dendritic cells (DC) and macrophages play a critical role in the initiation and maintenance of a T cell-mediated immune response. APC are essential for T cell priming, differentiation and activation in lymphoid tissue and at sites of inflammation. Understanding the molecular mechanisms via which APC function can be controlled may give novel insights into how T cell mediated immunity may be induced or blocked. NCRM (NIH Center for Regenerative Medicine) Lecture Thomas Graf became interested in the effects of genes on differentiation early on in his career. Originally a virologist, he co-discovered several avian acute leukemia oncogenes (myc, myb and erb) and developed in vitro cell transformation systems that exhibited the same lineage specificity as in the animal. He showed that a differentiation block is an essential feature of leukemogenesis and that at least two oncogenes need to cooperate to cause acute leukemia. Using a surrogate adult stem cell system, which he developed with leukemic avian cells, he was able to reprogram committed erythroid and myeloid cells first with signal transducing agents and later with lineage specific transcription factors. Extraordinary opportunities to study the molecular mechanisms that cause inherited diseases are being provided by new methods of producing stem cells. These techniques make it possible to produce from a patient cells that are equivalent to those early in their life. In principle many inherited diseases may be studied in this way including Motor Neuron Disease, schizophrenia, some forms of cancer and causes of sudden heart failure. In addition, human cells in the laboratory may provide important new approaches to the safety testing of new drugs.
